Methylcyclopropene-PEG4-NHS is a heterobifunctional linker containing a methylcyclopropene moiety for inverse electron demand Diels-Alder cycloaddition reactions and a NHS activated ester. The development of metabolic oligosaccharide engineering (MOE) as an important tool to visualize glycoconjugates in vitro and in vivo has significantly advanced glycobiology research during the past decade. In this approach, cells are grown in the presence of non-natural monosaccharides that contain a functional group with a unique reactivity (a chemical reporter), but are nevertheless accepted by the cell’s biosynthetic machinery. A carbamate-linked methylcyclopropene tag that is small enough to be accepted by the cell’s metabolism and at the same time highly reactive in the inverse-electron-demand Diels-Alder reaction (IEDDA).
