PTAD-alkyne is a heterobifunctional linker containing a PTAD moiety for tyrosine click reaction and an alkyne for click chemistry.
4-Phenyl-3H-1,2,4-triazoline-3,5(4H)-diones (PTADs) is a recently reported bioorthogonal probe that is capable of click reactions with tyrosine. Their benefits include but not limited to:
- Demonstrated selectivity against tyrosine over cysteine and lysine.
- Chemoselective for the phenolic side chain of tyrosine residues in modification of small molecules, peptides, and proteins
- Applicable in a wide variety of biologically acceptable aqueous buffers over a broad range of pH without using heavy metal catalysts.
- The tyrosine click linkage offers better stability than maleimide to temperature, extremes of pH, and exposure to human blood plasma.
